COMPARISON OF 14C-ACTIVITY DISTRIBUTION AFTER 14C-FLUORANTHENE AND 14C-GLYCINE UPTAKE BY YOUNG PEA PLANTS

The aim of this study was comparison of root and foliar uptake of 14C-fluoranthene (14C-FLT) and 14C-glycine (14C-Gly) to assess its translocation and distribution in different parts of young pea seedlings (Pisum sativum L.) and plants with two developed leaves. Transport and distribution of 14C-FLT (radioactive conc. 850 kBq/l, spec. activity 50 mCi/mmol) and/or 14C-Gly (radioactive conc. 1.7 MBq/l, spec. activity 53 mCi/ mmol) was monitored in pea seedlings that were colonized by aphids (Myzus persicae Sulzer). Monitoring was done during 72 hours of cultivation after uptake from the Richter’s nutrient solution via roots or spot application on the second developed leaf. After 2, 24, 48 and 72 hours distribution of 14C-activity in the root, epicotyl, cotyledons, second leaf, shoot apex and aphids was measured by liquid scintillation technique in dried samples after combustion in an oxygen flow. The accumulation of 14C-activity after both FLT and Gly uptake by pea seedlings from the nutrient solution was pronounced in both the root tip and the root differentiation zone compared to other plant parts. Significant 14C-activity was also detected in aphids. Plants with two developed leaves changed the distribution of 14C-activity, a significant accumulation of 14C-activity was also determined in the shoot apex. Aphids only moved from epicotyl to nodes with the first and second primary scales and to the apex region with undeveloped leaves. After spot treatment with the labelled substances on the second developed leaf, the maximum 14C-activity after FLT uptake in the aphids was detected as early in 24 hours, but in the case of Gly up to 72 hours after application. The results indicate a faster transport of FLT after absorption by roots compared to spot application on the leaf.
